The study of the behavior of animals under human care, elucidates answers that help in the conservation of species in natural environments. The analysis of reproductive behavior is capable of promoting the best methods for reproduction, conditioning and subsequent release of the offspring, in an attempt to restore environmental balance. The present study was carried out in a scientific breeding site for conservation purposes in Parque Vida Cerrado (Parque Fioravante Galvani), located in the rural area of the municipality of Barreiras, state of Bahia, Brazil. A sub-adult female maned wolf (Chrysocyon brachyurus) was observed for nine months, between August 2019 and March 2020, including the beginning of the fertile period, using the sequence sampling method to describe the behaviors observed, which is based on the record of a focal sample, allowing the standardization of methods for the elaboration of a behavioral repertoire, also making it important to report fortuitous, rare or unexpected behaviors. Cytological analysis, as complementary data, with the authorization of the responsible technician, were performed, totaling seven samples. The cytological data matched the behavioral data of each phase of the estrous cycle, allowing inferring in which phase of the estrous cycle the female accepts the male's charge and copulation, confirmed by spermatozoa in the cytological smears. The behavior data emitted in each estrous cycle are summarized by the female's sociability and tolerance of the male's permanence in its territory, inferring that the natural behavior of the species was not altered in the breeding site.
